I am preparing to venture to an apparently uninhabited island I saw on a map and was wondering if, instead of having a building, I could use a lighthouse as my main building to construct machinery in an such.

This would be useful only if I could build extentions/cottages/shacks inside... Is this possible?

I use a lighthouse to grow herbs but never tried to add any extension.

You can, I've got a library with an archive and a reading room inside a lighthouse. It used to be a prison, though... a dictator made my char build the cell when he was young.
